NCG-MHC-dKO mouse is an advanced immunodeficient model derived from the NCG strain (NOD/ShiLtJGpt background with Prkdc and IL2rg gene knockouts) and further engineered to lack both MHC class I and II molecules. The base NCG model already presents with severe combined immunodeficiency (lacking functional T cells, B cells, and NK cells) due to disrupted VDJ recombination (SCID mice) and impaired cytokine signaling (IL2rγ knockout). This results in loss of cellular and humoral immunity and makes the mice highly permissive for engraftment of human cells and tissues.
To further reduce immune rejection and minimize graft-versus-host disease (GvHD), MHC class I and class II genes (H2 complex) were knocked out using gene editing technology. MHC class I molecules, present on almost all nucleated cells, are essential for CD8⁺ T cell-mediated cytotoxicity and antigen presentation of intracellular pathogens or tumor antigens. MHC class II molecules, primarily expressed on antigen-presenting cells (APCs), are critical for initiating CD4⁺ T cell responses and play a central role in coordinating humoral and cellular immunity. The elimination of MHC molecules in NCG-MHC-dKO mice eliminates host-versus-graft and graft-versus-host alloreactivity, thereby:
- Extending the experimental window for human immune cell reconstitution, e.g. Human PBMC engraftment
- Stabilizing immune cell ratios,
- Improving accuracy in evaluating human antibodies and immunotherapies.
