T-cell engagers (TCEs) represent a cutting-edge class of immunotherapies that redirect a patient’s own T-cells to recognize and eliminate tumor cells by simultaneously binding to CD3 on T-cells and tumor-specific antigens. The development of TCEs poses unique preclinical challenges, including the necessity for a functional human immune system and the potential for adverse effects such as cytokine release syndrome. To address these challenges, specialized preclinical models that accurately mimic human immune-tumor interactions are essential.
